Abstract: Objective: To study the effects of Astragalus polysaccharides on the induction of inducible nitric oxide synthase (iNOS) induced by lipopolysaccharide (LPS) in human thyroid cells in vitro. Methods: Thyrocytes obtained from paraadenomatous tissue of thyroid adenomas were cultured in a monolayer system. Human thyrocytes were treated with LPS at different concentration (0, 1, 10, 100μg • ml-1) and at the same concentration for different time (6, 12, 18, 24 h). The level of nitric oxide (NO) in the culture medium was observed; Effect of iNOS inhibitor L-NAME on the above-mentioned effects of LPS; Effect of Astragalus polysaccharides on the above-mentioned effects of LPS. Results: (1) LPS (0, 1, 10, 100μg•ml-1) significantly increased the level of NO in dose-dependent manner. (2) LPS (6, 12, 18, 24 h) significantly increased the level of NO in time-dependent manner at the same concentration. (3) L-NAME inhibited the above-mentioned effects of LPS. (4) Astragalus polysaccharides inhibited the above-mentioned effects of LPS. Conclusions: Astragalus polysaccharides inhibited LPS-induced NO synthesis in iNOS in thyroid cells. Abstract: Objective: To investigate the effectiveness and precautions of shockwave-Intravascular Lithotripsy in severe calcified coronary artery disease. Methods: Three cases of severe calcified coronary artery disease treated with shockwave-Intravascular Lithotripsy were reviewed, and the literature on the application of shockwave- Intravascular Lithotripsy in coronary interventional therapy was systematically reviewed. Results: Severe calcified coronary artery lesions were found in all 3 patients by coronary angiography, and in 2 of them, intracoronary optical coherence imaging was used to further evaluate the location, type, and severity of calcification. All the 3 patients were treated with a shockwave-Intravascular Lithotripsy balloon catheter to help complete the pre-treatment of calcification lesions, and finally successfully implanted stents, achieving clinical success in coronary interventional therapy. Abstract: Objective: To investigate the relationship between hypoxia regulation-related lncRNA and the prognosis and treatment of patients with bladder urothelial carcinoma, so as to provide new ideas for clinical medicine. Methods: Clinical and genomic data were obtained from The Cancer Genome Atlas (TCGA) database, and hypoxia-related genes were obtained from the (GSEA) database. Prognostic models were constructed by co-expression analysis and Cox regression analysis. The roles of hypoxia-related lncRNAs in the tumor immune environment were revealed by GO, KEGG and tumor mutational burden (TMB) analysis. Results: A prognostic model containing 6 hypoxia-related lncRNAs was constructed. We found that low-risk patients had better overall survival (OS) and progression-free survival (PFS). ROC, independent predictive analysis, C-index, and nomogram indicated that hypoxia-related lncRNA models could independently predict patient outcomes. TMB analysis suggested that the biological function of hypoxia-related lncRNA was related to tumor immunity, and patients with low risk and high TMB had better prognosis.
